I have some 1/4" lucite ES acrylic, and was wondering if anyone has used this for an insert.. Being 1/4" it does have some give to it, but if I keep the insert size small, it may be OK to use.. I figure the router base would help stiffen it.. Any comments?

In my humble opinion, too thin. The weight of the router, the force the bit puts in the work, you'll have a wavy or inconsistent depth of cut, if it doesn't crack.

Of course, you can make a snazzy router compass, bit guards, and lots of other accessories with it.

1/4" lucite is not a good idea for two reasons: flex which has been mentioned, plus lucite has a tendency to shatter. Don't think I would want to deal with a shattered insert & a snarling router dropping down anywhere near my legs.
